Kurzdefinition

Cannflavine (Cannflavin A, B, C) sind prenylierte Flavone, die ausschließlich in Cannabis sativa vorkommen und durch ihre duale COX/LOX-Hemmung eine bis zu 30-mal stärkere anti-inflammatorische Wirkung als Aspirin entfalten.

Wissenschaftliche Beschreibung

Strukturelle Besonderheiten: Cannflavine sind 6-C-prenylierte Derivate von Luteolin (3',4',5,7-Tetrahydroxyflavon): - Cannflavin A: 6-(3,7-Dimethylocta-2,6-dien-1-yl)luteolin (Geranylgruppe an C-6); CAS 76735-57-4; Molekulargewicht 436,50 g/mol - Cannflavin B: 6-(3-Methylbut-2-en-1-yl)luteolin (Prenylgruppe an C-6) - Cannflavin C: methyliertes Derivat (8-Prenyl-Kämpferol-Typ), erst kürzlich charakterisiert

Biosynthese-Weg: Luteolin (aus Phenylpropanoid-Weg: p-Cumaroyl-CoA → CHS/CHI → Apigenin → F3'H → Luteolin) → [CsPT3, aromatische Prenyltransferase] → Luteolin + DMAPP (C₅) → Cannflavin B; Luteolin + GPP (C₁₀) → Cannflavin A → [CsOMT21, O-Methyltransferase] → methylierte Varianten.

Rea et al. 2019 (PMID 31151063): Rea und Kollegen identifizierten CsPT3 als die Prenyltransferase der Cannflavin-Biosynthese durch heterologe Expression in Tabak und In-vitro-Assays. CsOMT21 wurde als komplementäres Enzym für die Methylierung identifiziert. Der Biosyntheseweg wurde damit vollständig aufgeklärt.

Quantitativer Gehalt in Cannabis: - Cannflavin A: 0,14–0,40 % TG (Trockengewicht) in Blüten - Cannflavin B: 0,10–0,25 % TG - Cannflavin C: Spurenmengen (<0,02 % TG) - Gesamtcannflavin: 0,14–0,54 % TG; 5–10-fach höher in Trichom-Isolaten vs. Blattmaterial

Die erste pharmakologische Beschreibung stammt von Baxter & Williamson (1983, PMID 6271596), die zeigten, dass Cannabis-Extrakte stärker COX hemmen als erwartet – Cannflavine wurden als Wirkprinzip postuliert.

Neuroprotektive Aktivität (Eggers et al. 2019, DOI 10.1016/j.bcp.2019.08.011): Cannflavin A zeigt in PC12-Zellen einen hormetischen Effekt – bei Konzentrationen <10 µM Widerstandskraftsteigerung um 40%, Aβ1-42-Fibrillierungshemmung und Neuroprotektion gegen β-Amyloid-Toxizität; bei >10 µM Neurotoxizität. TEM-Auswertungen bestätigen reduzierte Aβ-Aggregat-Dichte.

Biologische Auswirkungen

  • Herbivoren-Abwehr: Starke COX/LOX-Hemmung hemmt Prostaglandin- und Leukotrien-Produktion in Insekten → insektizides Potential.
  • UV-Schutz: Aromatische Flavon-Chromophore absorbieren UV-Strahlung (λmax ~340–350 nm) → Schutz des Chloroplast-Apparats.
  • Antioxidativ: Catecholstruktur (3',4'-OH) ermöglicht radikalische Wasserstoff-Abgabe; DPPH-IC50 ~3–8 µM.
  • Antimikrobiell: Gehemmte LOX/COX-Wege stören Pilzinfektions-Signalwege; synergistische Wirkung mit Cannabinoiden gegen Botrytis (Hypothese).
  • Anti-amyloid: Direkte Hemmung von Aβ1-42-Fibrillierung in-vitro (Eggers 2019); potenziell relevant für Alzheimer-Pathogenese.

Pharmakologische Relevanz

Cannflavine sind wichtige Marker für die pharmazeutische Bewertung von Cannabisextrakten. Die duale COX/LOX-Hemmung überlagert sich nicht nur anti-inflammatorisch, sondern auch neuroprotektiv (Aβ-Aggregationshemmung). Der KMO-Effekt (IC50 29,4 µM) deutet auf eine zusätzliche metabolische Wirkung auf den Tryptophan-Katabolismus hin. In-vitro-Studien zeigen eine Dosis-Abhängigkeit mit optimalem Fenster <10 µM; höhere Konzentrationen werden neurotoxisch.

Chemotaxonomische Einordnung

Prenylierte Flavone mit C-6-Geranylierung sind in der Pflanzenwelt selten. Die Kombination aus CsPT3-Spezifität und CsOMT21-Aktivität erklärt die Cannabis-Exklusivität. In anderen Pflanzen (z.B. Mimulus, Diplacanthus) finden sich prenylierte Flavone, aber mit anderen Substitutionsmustern und ohne die spezifische biologische Aktivität der Cannflavine.

Analytik und Qualitätssicherung

LC-MS/MS ist der Standard für Cannflavin-Quantifizierung; GC-MS zersetzt die Glucoside-Variante. Referenzstandards für Cannflavin A/B sind kommerziell verfügbar (MedChemExpress). Detektion über charakteristische [M-H]⁻-Ionen und UV-Maxima bei ~340–350 nm.

Anbau-Relevanz

  • UV-B-Induktion: UV-B-Stressbehandlung (280–315 nm) steigert die Cannflavin-Synthese via CsMYB-Transkriptionsfaktoren und PAL-Induktion; 15–30 min UV-B täglich in der Spätblüte kann Cannflavin-Gehalte erhöhen.
  • Ernte und Extraktion: Cannflavine sind thermostabil (<150 °C); Alkohol-Extraktion (EtOH/MeOH) erschließt sie gut. CO₂-Extraktion (SFE) erfasst Cannflavine weniger effizient.
  • Züchtung: Hoch-Cannflavin-Sorten könnten durch Selektion auf CsPT3-Expressionslevel oder Markerassistierte Selektion (MAS) gezüchtet werden.
  • Sortenmarker: Cannflavin A/B-Verhältnis als potenzieller chemotaxonomischer Marker für verschiedene Chemovare.
