Photooxidativer Stress
Kurzdefinition
Photooxidativer Stress entsteht wenn photosynthetische Elektronentransportkette überreduziert wird: PSI → Mehler-Reaktion → O₂⁻/H₂O₂; PSII → ¹O₂ via Ladungsrekombination. Das Ascorbat-Glutathion-System (Halliwell-Asada) scavengert H₂O₂ im Chloroplast. Flavodiiron-Proteine (FLV) sind ein alternativer Elektronenabfluss ohne ROS-Intermediat (abwesend in Angiospermen, aber transgenes Potential).

H₂O₂ ist das stabilste ROS (~1 ms Halbwertszeit) und kann via Aquaporine zell-zu-zell transportiert werden → systemisches Signal.
Halliwell-Asada-Zyklus (Chloroplast)
APX (Ascorbat-Peroxidase): H₂O₂ + Ascorbat → 2 H₂O + MDHA
MDHAR (Monodehydroascorbat-Reduktase): MDHA + NADPH → Ascorbat + NADP⁺
DHAR (Dehydroascorbat-Reduktase): DHA + 2 GSH → Ascorbat + GSSG
GR (Glutathion-Reduktase): GSSG + NADPH → 2 GSH + NADP⁺
Ascorbat im Chloroplast: bis 20 mM (höchste Konzentration im Chloroplast = primärer H₂O₂-Puffer).
Chloroplast-Verlagerung
Hochlicht → Phototropin phot1/phot2 → Chloroplasten migrieren zu Antiklinalen Wänden → reduzierte Lichtabsorptionsfläche (Vermeidungsposition). Schwachlicht → Chloroplasten an Periklinalen Wänden → maximale Lichtabsorption (Akkumulationsposition).
Flavodiiron-Proteine (FLV)
FLV (FLV1-4) reduzieren O₂ direkt zu H₂O ohne O₂⁻- oder H₂O₂-Intermediat → sicherer alternativer PSI-Elektronenabfluss. In Angiospermen (inkl. Cannabis) von Natur aus abwesend (nur in Moosen, Algen, Cyanobakterien). Transgene FLV2/4 in Arabidopsis + Tabak: ↑ Toleranz gegen Hochlicht, Salz, Oxidantien (Vicino 2021). Potentielles Züchtungsziel für Cannabis unter intensiven LED-Beleuchtungssystemen.
Anbau-Relevanz
- LED-Hochleistungsanlagen: PPFD >1500 µmol/m²/s erhöht Photooxidations-Risiko; CO₂-Supplementierung auf >800 ppm senkt PSI-Überreduktion (Calvin-Zyklus läuft schneller)
- Ascorbat/Glutathion-Status: Nährstoffmangel (v.a. Schwefel für GSH-Biosynthese) → geschwächtes Antioxidantien-System → erhöhte Photooxidationsempfindlichkeit
- TBARS-Monitoring: Malondialdehyd-Gehalt (TBARS) als einfaches Biomarker-Assay für oxidativen Schaden
